چکیده
IntRoductIon Leprosy caused by Mycobacterium leprae (M. leprae), is a chronic granulomatous disease affecting the skin and peripheral nervous system, which is transmitted through direct contact with nontreated or inadequate treatment patients.[1] Diagnosis of leprosy depends on the clinical signs and symptoms and slit skin smear positivity. However, it’s sometimes similar with other granulomatous disease caused by mycobacterial infection. Early stage leprosy is difficult to diagnose by clinical criterion alone because the sensitivity of acid‐fast bacilli staining is quite low. The polymerase chain reaction‐restriction fragment length polymorphism (PCR‐RFLP) shows the great advantage in rapid identification and diagnosis for early cases and has a differentiation between leprosy and nonleprosy cases.

A RAPID AND SIMPLE METHOD OF DNA EXTRACTION FROM CLINICAL SPECIMENS CONTAINING MYCOBACTERIUM LEPRAE FOR PCR ASSAYS
We describe a rapid, simple, and reliable procedure for routine isolation of Mycobacterium leprae DNA from slit-skin swab specimens. This one-step DNA extraction method is based on the utilization of ChelexR 100, a chelating ion exchange resin. Slit-skin swab specimens from seven untreated leprosy patients at Baba Baghi Leprosy Hospital in Iran were processed by this procedure. The polymer...

In this article, we report on 2 indigenous cases of leprosy detected in a European country. We also report on the use of polymerase chain reaction-restriction fragment length polymorphism analysis of hsp65 gene for rapid identification of Mycobacterium leprae directly from the clinical sample.
متن کاملMolecular analysis of the S1 gene of vaccine strains of infectious bronchitis virus using reverse transcriptase-polymerase chain reaction and restriction fragment length polymorphism
Infectious bronchitis virus (IBV) is an acute and contagious viral disease of poultry that affects different systems, including the respiratory tract in particular. IBV causes major economic losses in the poultry industry globally. Due to antigenic variation of the causative agent, control of the disease is difficult. To control the disease, many vaccines that belong to different serotypes are...
متن کاملGenetic diversity of Staphylocoagulase genes (Coa) among methicillin-resistant Staphylococcus aureus isolates at clinical specimens of blood and urinary infections
Background: Staphylococcus aureus is a common pathogen in human that can be the cause of a wide range of infectious diseases including bacteremia, pneumonia, cellulitis, and osteomyelitis and skin and soft tissue infections. The coagulase enzyme is one of the most important virulence factors of this bacterium. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) Coa...
متن کاملDetection of avian reoviruses causing tenosynovitis in breeder flocks in Iran by reverse transcription-polymerase chain reaction (RT-PCR) and restriction enzyme fragment length polymorphism (RFLP)
BACKGROUND:Avian reoviruses (ARVs) are members of theOrthoreovirus genus; one of the 12 genera of the Reoviridaefamily. The ARVs are the cause of some important diseases inpoultry such as reovirus-induced arthritis, tenosynovitis, chronic respiratory disease, and mal-absorption syndrome.OBJECTIVES: In this study, the presence of ARVs in the Iranianbreeder flocks was investigated through reverse ...
